Summary |
The current study genotyped two variants of HTR1D gene in 272 ADHD trios of Chinese ethnicity, that is 1350T>C in the coding region and 1236A>G in 3'-UTR by the use of transmission disequilibrium test (TDT). The A allele of the 1236A>G polymorphism exhibited both a trend toward preferential transmission to ADHD probands and a significant preferential transmission to probands of ADHDC. Additional polymorphisms in this gene need to be studied further. |

Two hundred seventy-two nuclear families participated in the present study. ADHD probands were recruited from the ADHD outpatient clinic at the Child and Adolescent Psychiatric Division of the Sixth Hospital, Peking University in Beijing, PRC. |

Diagnosis Description |
All probands were interviewed by at least two different child psychiatrists with the aid of their biological parents and teacher and fulfilled DSM-IV diagnostic criteria for ADHD [USAn Psychiatric Association, 1994]. One hundred nine (40.0%) probands met criteria for ADHD combined type (ADHD-C), 17 (6.3%) met criteria for ADHD hyperactive-impulsive type (ADHD-HI), and 146 (53.7%) met criteria for ADHD inattentive type (ADHD-I). |
Technique |
Genomic DNA was extracted from whole blood by standard procedures. 1350T>C polymorphism was genotyped using the conditions described by Ozaki et al. [1995]. 1236A>G polymorphism was amplified with forward primer 5'-TCC TGT AAC CTC ATC GGG ATT G-3' and backward primer 5'-CGA GAG GGC AAG ACT CCG TC-3'. After initial denaturation at 95oC for 4 min, 35 cycles were carried out at 96oC for 30 sec, 53oC for 30 sec, and 72oC for 30 sec, followed by a final step of elongation at 72oC for 7 min. PCR products (10 ml) were digested with five units of EcoRV (MBI Fermentas, Vilnius, Lithuania) in 16 ml reaction volumes for 12 hr at 37oC. The digested PCR products were run on a 2.5% agarose gel at 100 V for 1.5 hr, then the agarose gel into water containing ethidium bromide for half an hour. Finally, they were analyzed with Gel Doc 2000 imaging system to detect and record the genotype of each sample. After digestion, the A variant was cut into two fragments of 66 and 170 bp, while the G variant was uncut and was 236 bp. Genotypes were read by two persons, both of whom were blind to the identity of the samples. |
